Barnsley will play League Two Morecambe in the EFL Cup first round on the week commencing August 7.
The two clubs have met just once before, with the Shrimps winning 2-0 at Oakwell in the same stage of the same cup in 2011.
Barnsley were knocked out of the cup by League One Northampton Town in the first round last season.
The league fixtures are due to be drawn on Wednesday.
